Los antibióticos betalactámicos (BL) son la causa más frecuente de alergia a fármacos, constituyendo como un 10% de la población general. Esto tiene enormes implicaciones ya que los facultativos se enfrentan a la decisión de elegir un tratamiento antibiótico alternativo, suponiendo más efectos secundarios y mayor coste. En la actualidad el desarrollo de nuevos antibióticos no es una prioridad para la industria farmacéutica y las tasas de resistencias bacterianas están en continuo aumento, por lo que la alergia a BL se ha convertido en un problema de salud global. Sin embargo, menos del 20% de los adultos y 10% de los niños considerados alérgicos lo son, por ello un correcto diagnóstico es clave para la seguridad del paciente y sostenibilidad del sistema sanitario.
La compleja reactividad química del clavulánico (CLV) ha dificultado la identificación de su determinante antigénico y por tanto se conoce muy poco sobre su inmunogenicidad ya que la respuesta alérgica al CLV implica generar conjugados de proteínas cuya estructura es inestable y de difícil identificación.
Para resolver la necesidad que se plantea, se han realizado diferentes aproximaciones sintéticas de determinantes potenciales del CLV, correspondientes a diferentes fragmentaciones del fármaco tras su unión a proteínas, para su respuesta inmunológica.

The most frequent reason of allergy to medicines is due to betalactam antibiotics (BL), resulting allergic an average 10% of the general population. This has enormous implications since the physicians face the decision to choose an alternative antibiotic treatment, which, though it is not their first choice, is more expensive and has more side effects.

At present, the development of new antibiotics is not a priority for the pharmacy industry meanwhile bacterial resistances rates are in continuous increase. Therefore, BL allergy has turned into a global health problem.

However, less than 20% of adults and 10% of children considered allergic to BL are indeed allergic. Due to this fact it is very important a correct diagnosis for the safety of the patient and sustainability of the sanitary system.

The complex chemical reactivity of clavulanic acid (CLV) has hindered the identification of its antigenic determinant and therefore very little is known about its immunogenicity. The allergic response to CLV involves generating protein conjugates whose structure is unstable and difficult to identify.

To solve the need that appears, different synthetic approaches of potential antigenic determinants of CLV have been made, corresponding to different fragmentations of the drug after its union to proteins, for its immune response.
